Vildagliptin is an orally administered dipeptidyl peptidase-IV (DPP-IV) inhibitor, following sitagliptin, approved by Novartis Pharmaceuticals Ltd. in 2008 for the treatment of type 2 diabetes in the European Union.
Vigliptin is a selective, competitive, reversible inhibitor of DPP24. Glucose-dependent  polypeptide (GIP) and glucagon-like polypeptide 21 (GLP21) are important hormones for maintaining glucose concentrations in the body, and both have enterostimulatory effects. the  effect of GIP is impaired in type 2 diabetes, and only GLP21 can exert effects, which promotes secretion by acting on receptors on the pancreatic β-cell membrane. GLP21 also inhibits the secretion of glucagon and inhibits gastric emptying, thereby increasing satiety (appetite suppression).

Gliptin is a highly selective and reversible substrate-like DPP-4 enzyme inhibitor, which has bidirectional regulation effects on blood glucose. Vildagliptin inhibits the activity of DPP-4 by combining with DPP-4 to form a DPP-4 complex, increases the concentration of active glucagon-like peptide-1 (GLP-1), and promotes the glucose-dependent growth of islet β cells. While secreting, it can also regulate α cells, and in the case of hyperglycemia, it can reduce glucagon levels. In addition, unlike other DPP-4 inhibitors, its inhibitory effect on DPP-4 is a substrate-like enzyme inhibition, so after fast binding to DPP-4, the dissociation is slower and the effect is longer.
