Orlistat is designed to treat obesity. Its primary function is preventing the absorption of fats from the human diet, thereby reducing caloric intake. Orlistat works by inhibiting pancreatic lipase, an enzyme that breaks down triglycerides in the intestine.

Function
Orlistat is a long-acting, powerful and specific gastrointestinal lipase inhibitor, which inactivates the above two enzymes by forming covalent bonds with lipase active serine sites in the stomach and small intestine. The inactivated enzymes cannot decompose the fats in food into free fatty acids and glycerol that can be absorbed by the human body, thus reducing fat intake and weight loss. In addition, it has been found that orlistat inhibits intestinal cholesterol absorption by inhibiting niemann-pickC1-like1 (NPC1L1).
Usage and Dosage
Orlistat combined with a slightly low caloric energy diet is indicated for the long-term treatment of obese and overweight patients including those who have developed obesity-related risk factors. Long-term weight control (weight loss, weight maintenance and weight loss prevention). It also reduces the incidence of obesity-related risk factors and other obesity-related diseases, including hypercholesterolemia and type 2 diabetes. For adults: The recommended dose is one 120mg capsule during or within one hour after a meal. If a meal is missed or the food does not contain fat, one dose of medication may be omitted. Long-term treatment effects, including weight control and improvement in risk factors, are sustainable. Patients should have a well-balanced diet with a low caloric energy, about 30% of which comes from fat, and a diet rich in fruits and vegetables. Fat, carbohydrate and protein intake should be divided into three meals a day. There is no evidence that more than 120mg three times a day enhances efficacy. Dose adjustment is not necessary for the elderly.
